Do you get two days front of line pass with a one night stay then?
you get the benefits from check in to the end of check out day
so, let's say you check in at 6:30am, you probably won't get your roombut you'll get your room key.
store your bags (if required) with bell services)
use your room key (even if you don't have the room yet) and print Express passes in the lobby. Insert your room key in the machine, it will take a picture of you, and issue an express pass (this is not your park ticket, you still need a park ticket)
head to the park with Early Entry, a Team Member will check your room key. Depending on when you go, it will be either IoA or USO that will have early admission. During the summer both parks have it.

Express passes are not available during early entry
as soon as the park opens for regular operations, you can use your express pass to enter the express lines.
when you're done for the day, or when you need a break, go back to the hotel (short walk or complimentary water taxi) to the hotel, you'll get your room after 4pm.
collect bags from bell services
the next morning store bags with bell services again, go to the parks for early admission, enjoy your express passes
when you're done, go back to the hotel, collect your belongings from bell services, and head to your next hotel/destination
An alternative scenario many here use,is to check in directly at your main destination (disney for example to take advantage of a free dining deal) then remain checked in at disney's for the whole duration
then at some point book a night at RPR for express (double booking, you pay for the room at universal as well as the room at disney's)
it still works cheaper than buying the express on the side
the main benefit is that, if you have a free dining, that's meals taken care of for more days (you stay at disney's all the while)
other benefits include not needing to pack up and haul suitcases (you can settle in at one hotel for the whole duration and just take a day bag to universal) and another benefit is that you do not need to decide if you're doing universal at the end of the stay or at the beginning, you can place your UOR night in the middle of your wdw stay, it's a nice change of resorts, but since you have a double booking, you don't loose your dining at disney's and you don't pack up. And it's still cheaper than doing the express passes on the side.
